FGFR3 alterations (bladder cancer)
FGFR3 is a growth-receptor gene mutated or fused in about a fifth of advanced bladder cancers and most low-grade early ones. It is the only targetable biomarker in bladder cancer so far.
FGFR3 point mutations (S249C, R248C, Y373C) and fusions (FGFR3-TACC3) define luminal-papillary tumours that are less immune-infiltrated. Erdafitinib is approved for FGFR3-altered disease after immunotherapy (THOR). Testing is by NGS on tissue or urine/ctDNA.
