MMAF is a charged cousin of MMAE that cannot cross membranes, so it kills only the targeted cell and spares neighbours. Eye side effects are characteristic.

Monomethyl auristatin F carries a charged C-terminal phenylalanine, which prevents the free payload from crossing membranes. Used with non-cleavable linkers (mafodotin), it has little bystander effect, less neuropathy than MMAE, and a distinctive corneal toxicity seen with belantamab mafodotin.
