Cell of origin (GCB vs ABC)

Whether a large B-cell lymphoma resembles a germinal-centre B cell or an activated B cell; the activated type does worse and depends on different pathways.

Determined by gene expression (Lymph2Cx) or the Hans IHC algorithm. ABC/non-GCB depends on NF-κB and BCR signalling (BTK, IRAK4, MYD88 L265P), GCB on BCL2/EZH2. Newer genetic classifications (LymphGen: MCD, BN2, N1, EZB, ST2, A53) refine this but are not yet clinically routine.
