Rindopepimut
A vaccine against a glioblastoma-specific mutant protein that looked promising for years and then failed its phase 3 trial in 2016. It is a landmark failure.
EGFRvIII peptide conjugated to KLH with GM-CSF. Phase 2 (ACT III) suggested prolonged survival versus historical controls; the double-blind phase 3 ACT IV (n=745, EGFRvIII-positive newly diagnosed glioblastoma with minimal residual disease) showed no OS benefit (20.1 vs 20.0 months). Loss of EGFRvIII expression at recurrence in both arms illustrated antigen escape. Celldex discontinued the programme.
